L'Afrique en 1915. Charlie Allnut, américain, transporte sur son bateau "l'African Queen" toutes sortes de marchandises qu'il distribue dans les villages. Il prévient le révérend Sawyer et sa soeur Rose, tous deux sujets britanniques, de l'approche des troupes allemandes. Il revient le lendemain et embarque Rose, pensant la déposer en territoire neutre. Mais, décidée à lutter, elle l'oblige à descendre les rapides du fleuve pour rejoindre les Anglais.
